摘要
采用濒危植物珙桐花药为外植体,研究了花序外形与小孢子发育时期的关系以及低温预处理、激素种类与浓度、蔗糖浓度等对诱导花药愈伤组织的影响并初步探讨了其诱导分化的可能性.结果表明:低温预处理对愈伤组织的形成影响不显著,随着低温时间的延长,愈伤组织诱导率逐渐降低;高浓度的蔗糖不利于愈伤组织的形成;愈伤继代后褐化严重未产生分化.在本实验中,诱导珙桐花药愈伤组织的最佳条件为:选用长度为1.7~2.2 cm的花序,4℃预处理2 d,培养基为MS+2,4-D 1 mg/L+NAA 0.5 mg/L+GA_31 mg/L+5%蔗糖,最高诱导率为91.3%.
The anthers of the endangered plant Davidia involucrata were chosed as explants for the first time to induce anther callus in this study. The relationship between bud appearances and developmental stages of microspores was identified first. And the anthers were cultured in MS mediums contain different hormone types, concentrations and sugar concentrations. The result indicated that the effect of lowtemperature pretreatment on callus is not significant. The callus inductivity is decreased gradually with the extending of low-temperature treatment time. It also indicated that high concentration of sugar is bad for the formation of callus. The best medium to induce anther callus in this study is using 1.7-2.2 cm long bud, 4 ℃ pretreated 2 day and the medium MS+2,4-D 1 mg/L+NAA 0.5 mg/L+GA3 1mg/L +5 % sugar; the highest inductivity is 91.3. After the callus induction, some explorations of the induction of bud had been done. However, due to the serious browning of callus, the differentiation was unsuccessful.
