摘要
目的:建立高效液相色谱(HPLC)外标法快速测定人血清中苯巴比妥的浓度。方法:血清样品经甲醇沉淀后,以甲醇-水(40∶60,V∶V)为流动相,流速为1.0 mL·min-1,色谱柱为Inertsil ODS-SP(150 mm×4.6 mm,5μm),柱温为30℃,紫外检测波长为214 nm。结果:血清内源性杂质不干扰待测物的测定,苯巴比妥的线性范围为4.5~72.8μg·mL-1,日内、日间精密度(RSD)均<5%,样品稳定性良好。结论:该法灵敏、快速、准确、操作简便、线性范围宽,可用于苯巴比妥的血药浓度监测。
ABSTRACT OBJECTIVE:To establish a HPLC external standard method human serum. METHODS: Serum samples were precipitated by methanol for rapid determination of Phenobarbital in followed by separation on the column of Inertsil ODS-SP ( 150 mm x4. 6 mm,5 I&m) at 30 ℃. The mobile phase consisted of methanol-water (40: 60,V: V) at a flow rate of 1.0 mL. min-l. The detecting wavelength was set at 214 nm. RESULTS: The intrinsic impurity of plasma did not interfere with the determination of analyte. The linear range of arbamazepine was 4. 5-72. 8 μg mL^-1. The inter-day and intra-day precisions were less than 5%. The samples showed good stability. CONCLUSION: The established HPLC external standard method is sensitive, fast, accurate and simple with wide linear range and thus it can be applied to the monitoring of serum concentration of Phenobarbital.
