摘要
目的:为了制备高效的,特异的抗人喉癌/抗蓖麻毒素A双功能单克隆抗体,观察其对Hep-2细胞株杀伤作用。方法:选用BALB/C鼠,分别行蓖麻毒素A和人喉鳞状细胞癌腹腔免疫后,提取抗体,经与骨髓瘤细胞融合,制备成抗人喉癌/抗蓖麻毒素A双特异性单克隆抗体。结果:该双功能抗体与喉癌组织的特异性结合率为87%,对Hep-2细胞半数抑制浓度比游离环磷酰胺强40倍。结论:采用二次杂交瘤技术制备的双功能抗体优于化学偶联法,抗体效价具有均匀性和理想的信噪比,抗体易于标准化。
To investigate the preparation of a human laryngeal cancerbispecific antibody. Methods: The second set hybrid tumor technology was applied to prepare an anti-human laryngeal cancer and anti-castor toxin A bispecific monoclonal antibody. Results: The specific conjugation rate between bispecific monoclonal antibody and laryngeal cancer tissue was 87%. The half inhibition concentration to Hep-2 cell was 40 times stronger than that of free cyclophosphamide. Conclusion:The second set hybrid tumor technology to prepare the bispecific antibody had a homogeneous titer,an idealsignal-to-noise ratio and was easy to be standardized.
