摘要 目的:分析不同采收途径及不同部位的白花丹中白花丹醌含量,寻找富集白花丹醌的药用部位和白花丹的最佳采收途径。方法:采用HPLC法测定不同采收途径白花丹和不同部位中白花丹醌的含量:色谱柱为Hypersil ODS(25cm×4.6mm,25μm),流动相为甲醇-水(65∶35),流速为1.0mL/min,检测波长为213nm,柱温30℃。结果:白花丹醌在0.01μg~0.32μg含量范围呈线性关系,Y=18211600X+10434.57758,r=0.99946,RSD=1.28%(n=6);新鲜的白花丹根部、茎部及叶中白花丹醌的含量分别为0.026%、0.006%和0.014%;干燥的白花丹根部、茎部及叶中白花丹醌的含量分别为0.324%、0.082%和0.174%;枯萎的白花丹茎部、叶及穗中白花丹醌的含量分别为0.002%、0.001%和0.003%。结论:干燥组白花丹中白花丹醌含量高于新鲜组和枯萎组,白花丹根部的白花丹醌含量最高,其次是穗,叶、茎。本法简便、准确,灵敏度高,重复性好,可用于控制白花丹药材的质量。 Objective:to find the Plumbago Zeylanica medicinal materials with the high content plumbagin,to separately detect the Plumbagin content which come from different medicinal parts and different harvest ways of Plumbago Zeylanica growth in Nanning,Guangxi,to provide the basis for optimization of medicinal materials.Methods:the chromatographic column is Hypersil ODS(4.6mm×250mm,25μm),the mobile phase is methanol-water(65∶ 35),the flow rate is 1.0ml/min,the detection wavelength is 213nm,the column temperature is 30℃.Results:plumbagin has good linear relation within 0.01μg^0.32μg,and the regression equation isY=18211600X+10434.57758,r=0.99946(n=6),RSD=1.28%(n=6).The Plumbagin content of the fresh Plumbago Zeylanica medicinal materials roots,stems and leaves is respectively0.026%,0.006%,0.014%;the Plumbagin content of dried Plumbago Zeylanica medicinal materials roots,stems and leaves is respectively0.324%,0.082%,0.174%;the Plumbagin content of withered Plumbago Zeylanica medicinal materials stems,leaves and inforesences is respectively 0.002%,0.001%,0.003%.Conclusion:the plumbagin content of dried Plumbago Zeylanica medicinal materials group is higher than the fresh group and withered group,which withered group is the lowest;the highest Plumbagin content of each medicinal parts is the roots,followed by Inflorescences,leaves,stems were the lowest.The method is simple,accurate,sensitive,reproducible,and can be used to control the quality of Plumbago medicine.
