当归-牛膝对兔骨关节炎PI3K/AKT通路的影响 被引量:15

Effects of Angelica-Achyranthes on the PI3K/AKT Pathway in the Rabbit Knee Osteoarthritis

摘要 目的:为探讨当归注射液-牛膝总皂甙对兔膝骨关节炎软骨细胞凋亡及PI3K/AKT通路的影响。方法:60只新西兰兔随机分为6组,每组10只,分别为:正常组、模型组、当归注射液-牛膝总皂甙组、牛膝总皂甙组、布洛芬组、当归-牛膝中药颗粒配伍组,正常组常规饲养余各组采用改良Hulth法复制膝骨关节炎模型。取材前14d灌胃,造模术后45d处死。流式细胞术检测软骨细胞的凋亡率,应用免疫印迹法检测关节软骨PI3K/AKT通路AKT的Thr308表位活化程度。结果:①软骨细胞凋亡率检测:模型组软骨细胞凋亡率高于正常组;当归注射液-牛膝总皂甙组、牛膝总皂甙组、布洛芬组分别与模型组比较有统计学差异(P<0.01),组间比较无差异;中药颗粒组与模型组比较有统计学差异(P<0.01);中药颗粒组低于布洛芬组有统计学差异(P<0.05);当归注射液-牛膝总皂甙组和牛膝总皂甙组的凋亡率均低于当归-牛膝中药颗粒配伍组有统计学差异(P<0.05)。②软骨组织AKT1的表达量:模型组的软骨组织AKT的Thr308表位活化程度低于正常组,当归注射液-牛膝总皂甙组、牛膝总皂甙组和布洛芬组与正常组接近,中药颗粒组低于正常组。当归注射液-牛膝总皂甙组、牛膝总皂甙组和布洛芬组软骨组织AKT的Thr308表位活化程度高于模型组,中药颗粒组与模型组相近。当归注射液-牛膝总皂甙组、牛膝总皂甙组和布洛芬组软骨组织AKT的Thr308表位活化程度几近相等,以当归注射液-牛膝总皂甙组稍高,比中药颗粒组高。结论:当归注射液-牛膝总皂甙组及布洛芬组可以有效的减少骨关节炎病变过程中软骨细胞的凋亡,一定程度改善骨关节炎病理,其机制可能与其干预兔早期膝骨关节炎软骨组织PI3K/AKT信号通路有关。 Objective To study the role of Angelica injection-bidentata saponins on chondrocyte apoptosis and the PI3K/AKT pathway in the rabbit knee osteoarthritis.Methods 60 rabbits were randomly divided into 6 groups(n=10) were as follows: normal group,model group,Angelica injection-bidentata saponins group,bidentata saponins group,the ibuprofen group,Angelica-Achyranthes medicine particles compatibility group,normal group routine feeding,modified Hulth replicated knee osteoarthritis model.Drawn 14 days before the gavage,the modeling were sacrificed 45 days after surgery.Chondrocyte apoptosis rate by flow cytometry,Western blot technic of articular cartilage PI3K/AKT pathway AKT Thr308 epitope activation extent.Results ①Cartilage cells of a chondrocyte apoptosis detection: model group the rate of apoptosis in the normal group;Angelica injection-bidentata saponins group,bidentata saponins group,the ibuprofen group were statistically significant compared with model group(P〈0.01),but between the two groups no difference;Chinese medicine granules with model group,a statistically significant(P〈0.01);Chinese medicine granules group was lower than the ibuprofen group was statistically significant(P〈0.05);Angelica injection-bidentata saponins group and bidentata saponins group apoptosis rate was lower than Angelica-Achyranthes Chinese medicine granules compatibility groups are statistically significant(P〈0.05).② the amount of cartilage tissue AKT1 expression: the model group,the cartilage tissue AKT Thr308 table bit activation was lower than normal group,Angelica injection-bidentata saponins group,bidentata saponins groups and the ibuprofen group and normal group close to Chinese medicine granules group was lower than the normal group.Angelica injection-bidentata saponins group,bidentata saponins group and the ibuprofen group cartilage tissue AKT Thr308 table bit activation than the model group,similar to traditional Chinese medicine granules with the model group.Angelica injection-bidentata saponins group,bidentata saponins group and the ibuprofen group cartilage tissue AKT Thr308 table bit activation extent nearly equal to Angelica injection-bidentata saponins group is slightly higher than the Chinese medicine granules group.Conclusion Angelica injection-bidentata saponins group and the ibuprofen group can effectively reduce the apoptosis of chondrocytes in the osteoarthritis disease process,to a certain extent to improve the pathology of osteoarthritis,which may relate to intervention in osteoarthritis of the knee cartilage of rabbit early organization PI3K/AKT pathway.
