H_(615)可移植性肝癌碱性磷酸酶同工酶研究

摘要 本文研究了H615肝癌组织AKP同工酶的活性、电泳行为、热稳定性及对L-苯丙氨酸抑制的敏感性。H615肝癌与正常615鼠肝组织AKP酶活性分别为13.87±5.32单位/克组织及31.30±1.62单位/克组织,二者间有显著性差(P<0.01);在淀粉胶和聚丙烯酰胺凝胶电泳上,H615肝癌AKP与正常615小鼠肝、小肠、胎肝及胎盘AKP有不同的迁移率;此外,H615肝癌AKP比正常肝AKP热稳定性强而对L-苯丙氨酸抑制不敏感。H615肝癌AKP同工酶这些质和量上的变化提示H615肝癌组织具有一种特殊的AKP同工酶,这可能是该肿瘤细胞中一个低水平表达的异位基因的表达产物。 Alkaline phosphatase isoenzymes(E. C. 3. 1. 3. 1.)in a transplantable hepatoma of 615 mice(H_(615)AKP) were studied by means of measuring the enzyme activity, electrophoresis, thermostability and L—phenylalanine inhibition tests in this paper. The results showed that the enxymatic activities of H_(615) and normal liver AKP in 615 mice were 13.88±5.32 and 31.30±1.62 respectively with significant differnce statistically. On the starch—gel and polyacrylamide electrophoretic patterns, the eiectrophoretic mobility of H615 AKP was different from that of others tissue AKP including normal liver. fetal liver, small intestine and placente AKP in 615 mice. In addition, the thermostability of H_(615) AKP was higher than that of normal liver AKP and H_(615) AKP was insensitive to L—phenylalanine inhibition. The quantitative and qualitative bifference of H_(615) AKP from others normal AKP indicated that the hepatoma AKP was a specfic AKP isoenzyme and, perhaps, was an abnormal product of expression of a various AKP gene in the cancer cells..
