摘要
目的明确促卵泡发育的卵巢卵母细胞自分泌因子生长分化因子-9(GDF-9)、骨形态发生蛋白-15(BMP-15)的表达不足在免疫性卵巢早衰发病中的重要作用,进一步阐明补肾活血中药复方左归丸通过调节卵母细胞自分泌因子的表达,对卵巢起到保护作用。方法建立小鼠免疫性卵巢早衰模型:以小鼠透明带3所合成透明带多肽为免疫原。以出现动情周期紊乱、卵巢细胞受损,表示造模成功。然后将造模成功的小鼠随机分为模型组、左归丸高、低剂量组,强的松组、己烯雌酚组和对照组,共灌胃给药4周。免疫组化法检测GDF-9、BMP-15蛋白的表达。结果模型组与空白对照组相比,GDF-9蛋白表达水平下降,差异显著(P<0.05),左归丸低、高剂量组及己烯雌酚组与模型组相比,GDF-9蛋白表达水平增加,差异显著(P<0.05),与空白对照组相比,无显著差异。左归丸高剂量组GDF-9蛋白表达水平也较强的松组增加,差异显著(P<0.05)。模型组BMP-15蛋白表达水平较其余各组减少,但各组相比均无显著性差异(P>0.05)。结论左归丸对卵巢具有保护作用,能促进卵巢GDF-9蛋白的表达,从而促进卵泡的发育。
Objective To explore the effect of Zuogui pills(ZGP),a Chinese compound recipe for tonifying Shen(a term of Chinese traditional medicine for kidney),on the expression of GDF-9 in mice with autoimmune premature ovarian failure(APOF).Methods SPF BALB/C mouse models of APOF were established by subcutaneous injection of murine zona pellucida 3 at multiple sites.Then these mice were treated with various doses of ZGP(low,medium and high dosages),prednisone and diethylstilbestrol(DES) for 4 weeks respectively.The expression of GDF-9 was detected by immunohistochemical method.Results In comparison with control group,level of GDF-9 was significantly decreased(P0.05) in model group.The expression of GDF-9 was significantly increased in low dosage ZGP,high dosage ZGP and DES groups than that of model group(all P0.05).However,there was no significant difference among them compared with that of control group.In addition,the expression of GDF-9 was increased in high dosage ZGP group compared with prednisone group,and their difference was significant(P0.05).The expression level of BMP-15 protein in model group was decreased compared with other groups,but the difference between each group was not significant(P0.05).Conclusion ZGP may have a protective effect on ovary,and it can promote the expression of GDF-9 protein,which may contribute to the follicular development of ovary.
