4-羟雌二醇与肿瘤关系的研究进展

摘要 4-羟雌二醇(4-hydroxyestradiol,4-OHE2)是内源性雌激素代谢产物中活性最强的一种,属一种致癌物质。其代谢产物儿茶酚雌激素-3,4-苯醌(CE-3,4-Q)和在代谢过程中产生的氧自由基都具有基因毒性,可与DNA结合,导致DNA的损伤。4-OHE2可激活基质金属蛋白酶的前体(ProMMPs)生成基质金属蛋白酶(MMPs),促进肿瘤的转移与发展。4-OHE2可激活细胞核因子(NF-κB)参与细胞的信号转导,还可激活细胞外信号调节蛋白激酶(ERK)/丝裂原活化蛋白激酶(MAPK)路线导致肿瘤的发生,因此4-OHE2与肿瘤的发生发展有着密切的关系。
