低相对分子质量肝素对裸鼠胃癌模型CD40／CD40L表达的影响

Effect of low mloecular weight heparin on CD40/CD40L expression of orthotopic transplantation gastric tumor model in nude mice

目的观察低相对分子质量肝素（LMWH）对裸鼠胃癌模型CD40／CD40L表达的影响。方法应用原位移植方法建立裸鼠胃癌原位模型，术后7d将裸鼠模型随机分为2组：对照组、LMWH治疗组。每周注射药物2次，共5周，第6周处死裸鼠，观察两组裸鼠胃癌模型成瘤情况，采用免疫组织化学染色法检测肿瘤组织CD40／CD40L的表达。结果LWMH组胃癌原位模型肿瘤体积较对照组明显缩小，抑瘤率为42．25％，远处转移率为20％，较对照组70％显著降低（P〈0．05）；LMWH组CIMO表达水平显著低于对照组（P〈0．05），而CIMOL表达水平显著高于对照组（P〈0．05）。结论LMWH可抑制裸鼠胃癌的增殖及转移，其机制可能与干预肿瘤CD40／CIMOL通路有关。

Objective To study the effect of Low-molecular weight heparins on the expression of CD40/CD40L in orthotopic transplantation gastric tumor model in nude mice. Methods A model of human gastric cancer was established by orthotopic transplantation of histologically intact tumor tissue blocks in the gastric walls of nude mice. All mice were randomly divided into 2 groups ： control group and LMWH group. Forty-two days after treatment, tumor proliferation and metastasis rates were evaluated respectively, The expressions of CD40/CIMOL proteins were detected by immunohistochemistry. Results In comparison with that in the control group, the tumor volume of LMWH group was reduced significantly, and the inhibi- tory rate was 42. 25%. The distant metastasis rate （20%） compared with the control group （70%） was significantly lower （ P 〈 0. 05 ） ; The expressions of CD40 protein in the LMWH group was significantly low- er than that in the control group （ P 〈 0. 05）, The expressions of CD40L protein in the LMWH group was significantly higher （P 〈 0. 05 ）. Conclusion Low-molecular weight heparins has inhibitory effects on tumor proliferation and metastasis, the mechanism may interfere in CD40/CD40L passageway.
