摘要
目的观察低相对分子质量肝素(LMWH)对裸鼠胃癌模型CD40/CD40L表达的影响。方法应用原位移植方法建立裸鼠胃癌原位模型,术后7d将裸鼠模型随机分为2组:对照组、LMWH治疗组。每周注射药物2次,共5周,第6周处死裸鼠,观察两组裸鼠胃癌模型成瘤情况,采用免疫组织化学染色法检测肿瘤组织CD40/CD40L的表达。结果LWMH组胃癌原位模型肿瘤体积较对照组明显缩小,抑瘤率为42.25%,远处转移率为20%,较对照组70%显著降低(P〈0.05);LMWH组CIMO表达水平显著低于对照组(P〈0.05),而CIMOL表达水平显著高于对照组(P〈0.05)。结论LMWH可抑制裸鼠胃癌的增殖及转移,其机制可能与干预肿瘤CD40/CIMOL通路有关。
Objective To study the effect of Low-molecular weight heparins on the expression of CD40/CD40L in orthotopic transplantation gastric tumor model in nude mice. Methods A model of human gastric cancer was established by orthotopic transplantation of histologically intact tumor tissue blocks in the gastric walls of nude mice. All mice were randomly divided into 2 groups : control group and LMWH group. Forty-two days after treatment, tumor proliferation and metastasis rates were evaluated respectively, The expressions of CD40/CIMOL proteins were detected by immunohistochemistry. Results In comparison with that in the control group, the tumor volume of LMWH group was reduced significantly, and the inhibi- tory rate was 42. 25%. The distant metastasis rate (20%) compared with the control group (70%) was significantly lower ( P 〈 0. 05 ) ; The expressions of CD40 protein in the LMWH group was significantly low- er than that in the control group ( P 〈 0. 05), The expressions of CD40L protein in the LMWH group was significantly higher (P 〈 0. 05 ). Conclusion Low-molecular weight heparins has inhibitory effects on tumor proliferation and metastasis, the mechanism may interfere in CD40/CD40L passageway.
