桑白皮多糖提取工艺正交试验优化研究

摘要 目的优选桑白皮多糖最佳提取工艺。方法采用正交试验设计,以提取次数、提取时间及醇沉浓度作为工艺因素,每个因素采用3个水平,选择L9(34)正交表进行试验,同时用硫酸-苯酚法测定桑白皮多糖的含量作为评价指标。结果桑白皮多糖的最佳提取工艺为超声提取30 min,提取2次,醇沉浓度为80%。结论优选的工艺多糖提取率稳定,提取率高,提取工艺合理可行。 Objective To optimize the extraction process of polysaccharides of Cortex Mori.Methods Orthogonal experimental design was adopted,with extraction time,ultrasonic time and the concentration of alcohol as factors,3 levels in each factor,and L9(34) orthogonal table was used.The content of Cortex Mori polysaccharides was determined by sulfuric acid-phenol method as the index.Results The optimum extract process was as follows:ultrasonic extracted 30 min for two times,the concentration of alcohol was 80%.Conclusion This method is stable and efficient,the optimum process is reasonable and feasible.
