摘要
研究了二溴羟基苯基荧光酮 (DBHPF) Mo(Ⅵ )配合物作为蛋白质光谱探针的结果 .在pH 2 0~4 0HCl NaAc缓冲介质中 ,乳化剂OP存在下 ,DBHPF Mo(Ⅵ )室温下与蛋白质迅速形成稳定的复合物 ,使DBHPF Mo(Ⅵ )配合物 532nm处吸收峰值降低 ,根据这一变化可以定量测定微量蛋白质 .该法选择性佳 ,稳定性好 (达 2 4h) ,可直接用于人尿或人血清蛋白质的定量测定 。
The application of dibromohydroxyphenyl fluorone(DBHPF) Mo(Ⅵ) complex as a spectroscopic probe for protein is studied.In the buffer medium of HCl NaAc at 2 0~4 0 and in the presence of p octylpolyethylenther(OP) ,in ordinary temperature,DBHPF Mo(Ⅵ) complex combines protein rapidly to form a stable compound,which results in change of absorption spectrum of DBHPF Mo(Ⅵ) complex and decreases the absorbance peak value at 532?nm.According to these changes,the microdetermination of protein can be described.The method is selective,stable for at least 24?hours and can be used to directly determine protein in human urine and serum with same results as by Bradford method.This system can be applied to the clinic detection.
