清热益气中药对初发2型糖尿病KKay小鼠胰岛素敏感性的影响 被引量:8

Influence of medicinals with actions of clearing heat and tonifying qi on insulin sensitivity in KKay mice with early type 2 diabetes

摘要 目的观察清热益气中药干预初发2型糖尿病KKay小鼠血糖水平、胰岛素敏感指数的效果及肝脏病理形态变化,探讨中药治疗初发2型糖尿病的作用机制。方法 10周龄雄性初发2型糖尿病KKay小鼠27只按血糖值随机分为模型组、罗格列酮组[0.67 g/(kg.d)]、中药组[8.3 g/(kg.d)]。正常对照组选10周龄雄性C57BL/6J小鼠9只。各组连续灌胃给药4周后取材,测定其随机血糖、空腹血糖、胰岛素,肝脏切片HE染色观察组织学改变。结果给药4周后,与正常组相比,模型组胰岛素敏感指数明显降低(P<0.01),空腹血糖、随机血糖明显升高(P<0.01),肝脏组织明显脂肪变性。中药组与模型组相比,胰岛素敏感指数高于模型组(P<0.01);空腹血糖值低于模型组(P<0.01);中药组随机血糖值较模型组有下降,但无统计学意义(P>0.05);中药组肝脏结构明显好于模型组。结论清热益气中药可改善初发2型糖尿病KKay小鼠的胰岛素抵抗,增加胰岛素的敏感性从而改善血糖水平,并能有效保护肝脏组织形态结构。 Objective To observe the influence of medicinals with actions of clearing heat and tonifying qi on the level of plasma glucose, insulin sensitive index (ISI) and pathomorphology of liver in KKay mice with early type 2 diabetes. Methods Male KKay mice with early type 2 diabetes ( 10 weeks old, n = 27 ) were divided into model group, rosiglitazone group (0.67 g/kg · d) and Chinese medicinal group (8.3 g/kg · d) according to the value of plasma glucose. Other male C57BL/6J mice (I0 weeks old, n = 9) were selected as normal group. All groups were intragastrically given corresponding medicinals for d weeks and then the levels of random blood sugar (RBS), fasting plasma glucose (FPG) and isuline were detected. The histomorphologic changes of liver were observed by using HE staining of liver section. Results Compared with normal group, ISI decreased significantly (P 〈 0. 01 ), the levels of FPG and RBS increased significantly ( P 〈 0.01 ) and liver tissue had obvious adipose degeneration in model group. Compared with model group, ISI increased (P 〈 0.01 ), FPG decreased (P 〈 0.01 ) and RBS decreased but without statistical significance (P 〉 0. 05 ) in Chinese medicinal group. The histomorphologic structure of liver was better in Chinese medicinal group than that in model group. Conclusion Medicinals with actions of clearing heat and tonifying qi can alleviate the insulin resistance, improve insulin sensitivity, regulate the level of blood sugar and protect the histomorphologic structure of live in KKay mice with early type 2 diabetes.
