TLC法检测熊去氧胆酸中的胆石酸

Determination of lithocholic acid in Ursodeoxycholic acid by TLC

目的采用TLC法测定熊去氧胆酸中胆石酸的含量。方法采用硅胶G薄层板,以二氯甲烷-丙酮-冰醋酸(85∶5∶3)为展开剂,10%磷钼酸的硫酸-冰醋酸(1∶20)溶液为显色剂。结果 TLC法能有效检出熊去氧胆酸中的胆石酸,检测限为0.02μg。结论所用方法专属性好、灵敏度高,可用于熊去氧胆酸中胆石酸的检查。

OBJECTIVE To establish a method for the examination of lithocholic acid in ursodeoxycholic acid by TLC.METHODS Silica gel G was used as stationary phase and a mixture of dichloromethane,acetone and glacial acetic acid（85：5：3） was used as mobile phase.The color-developing reagent was 10% phosphomolybdic acid in concentrated sulfuric acid and glacial acetic acid solution（1：20）.RESULTS The method was proved effective to detect lithocholic acid and the detection limit was 0.02 μg.CONCLUSION This method is specific,sensitive and can be used to examine lithocholic acid in ursodeoxychlic acid.
