不同质量浓度的蔗糖和硼酸对北京杨花粉萌发的影响 被引量:12

摘要 以北京杨新鲜花粉为材料,研究固体培养基中蔗糖和硼酸不同质量浓度对花粉萌发的影响,结果表明:在培养基不含硼酸时,最适宜萌发的蔗糖质量浓度为10%,萌发率93.5%;当达到20%时,抑制花粉萌发,萌发率仅为55.6%。在培养基不含蔗糖时,最适宜萌发的硼酸质量浓度为0.01%,随着质量浓度升高,萌发率呈下降状态。在蔗糖和硼酸的双重作用下,最适宜萌发的质量浓度为10%蔗糖+0.01%硼酸,萌发率高达98.6%,花粉管生长快、健壮。新鲜花粉生活力测定的培养时间为24 h。 The effect of sucrose and boric acid with different concentration in solid medium on the pollen germination of Populus × beijingensis were studied in this paper.Results showed that without the boric acid,the optimal concentration of sucrose was 10%,and the pollen germination rated was 93.5%.The pollen germination was only 55.6% if the concentration of sucrose was 20%.Without sucrose,the optimal concentration of boric acid was 0.01%.The pollen germination rate decreased with the increasing concentration of boric acid.With both sucrose and boric acid,the optimal medium was with 10% sucrose and 0.01% boric acid.Under this condition,the pollen germination rate was 98.6%,and the pollen tube grew fast and healthy.The cultivating time for measuring viability of fresh pollen was 24 h.
