摘要
以籼稻品种特青的幼胚作为外植体 ,研究了激素对愈伤组织诱导及植株再生的影响 .MS+2 ,4 -D2 mg/ L作培养基 ,愈伤组织诱导率达 10 0 % ;愈伤组织在 MS+KT3mg/ L+NAA0 .5mg/ L的培养基上 ,分化频率为 6 7.3% ;在此基础上利用基因枪转化技术 ,将外源的 bar基因导入了水稻基因组 ,转基因植株表现出对除草剂 Basta很强的抗性 .
Using immature embryos of indica rice cultivar Teqing as explants, the effects of hommones on the induction of calli and the regeneration of plants were investigated. The induction frequency of calli on MS + 2,4\|D 2 mg/L medium was 100%; When MS + KT 3 mg/L + NAA 0.5 mg/L was used as differentiation medium,the differentiation frequency of buds reached as high as 67.3%. Immature embryos were bombarded with the plasmid pCB1 containing a \$bar\$ gene. Five transgenic plants were regenerated after selection. The result of PCR assay showed that foreign genes had been integrated into the rice genome. Transformed T0 plants showed high resistance to herbicide Basta.
