摘要
目的:建立同时测定虎杖中虎杖苷和白藜芦醇含量的方法,并对西南5个产区虎杖样品中2种成分的含量进行比较。方法:采用高效液相色谱法同时测定2种成分。色谱柱为AgilentC18(150mm×4.6mm,5μm),流动相为乙腈-水(23:77,V/V),检测波长为306nm。结果:虎杖苷和白藜芦醇的进样浓度分别在48~480μg.mL-1(r=0.9990)、7.2~72.0μg.mL-1(r=0.9992)范围内与各自峰面积积分值呈良好的线性关系;两者平均加样回收率分别为98.69%和99.42%,RSD分别为1.68%和3.39%(n均为6)。西南5个产区虎杖中虎杖苷含量高低依次为四川乐山>四川都江堰、重庆永川>贵州遵义、贵州毕节;白藜芦醇含量高低依次为贵州遵义、贵州毕节>四川都江堰、四川乐山>重庆永川。结论:本试验结果可为虎杖的质量控制提供参考依据。
OBJECTIVE:To establish the method for simultaneous determination of polydatin and resveratrol in Polygonum cuspidatum,and to compare 2 ingredients in samples from 5 different regions.METHODS:HPLC method was adopted to determine the contents of 2 components simultaneously.The analysis was achieved with an Agilent C 18(150 mm×4.6 mm,5 μm)column with acetonitrile-water(23:77,V/V).The detection wavelength was set at 306 nm.RESULTS:The linear ranges were 48~480 μg.mL-1 for polydatin(r=0.999 0) and 7.2~72.0 μg.mL-1 for resveratrol(r=0.999 2).The average recoveries were 98.69%(RSD= 1.68%,n=6)and 99.42%(RSD=3.39%,n=6).The contents of polydatin in samples from Leshan was higher than Dujiangyan of Sichuan province,that of Yongchuan of Chongqing city was higher than that of Zunyi and Bijie of Guizhou province;the contents of resveratrol in samples from Zunyi and Bijie of Guizhou province were higher than that of Dujiangyan of Sichuan province,that of Leshan of Sichuan province was higher than Yongchuan of Chongqing city.CONCLUSION:The study can provide reference for the quality control of P.cuspidatum.
