摘要
介绍一种用高效液相色谱 ( HPLC)快速准确测定大豆磷脂中主要活性成分磷脂酰胆碱( PC)的新方法。采用硅胶色谱柱 ,以正己烷 -异丙醇 -磷酸 -水为流动相 ,紫外检测器在 2 0 6 nm下检测 ,样品中 PC和其它磷脂组分完全分离 ,PC出峰时间短 ( <1 0 min)。样品经溶解后可直接进样 ,不需任何预处理。
A successful method for separation and quantitative determination of phosphotidylcholine (PC) in soybean lecithin by HPLC has been developed. The separation was performed on a licrosorb Si60 column, 250mm×4.6mm.i.d, by isocratic elution of nhexane∶2propanol∶phosphoric acid∶H_2O(45∶48∶0.15∶7.5,V∶V). The phopholipids were detected at 206nm. Under this condition, the retention time of PC is lower than 10min, the average recoveries of PC is 97.8%. The linear range of PC is between 0.05~4.00g/L and relative coefficent is 0.998. This method has been used to analysis PC in soybean lecithin from different sources.
