环黄芪醇对D-半乳糖致衰老小鼠的抗衰老作用 被引量:22

摘要 目的:探究环黄芪醇(cycloastragenol,CAG)延缓衰老作用及其机制。方法:采用D-半乳糖致小鼠衰老模型,小鼠随机分为正常组、模型组、环黄芪醇低、中、高3个剂量组(2.5,5.0,10.0 mg.kg-1)和维生素E(VE)组(2.5 mg.kg-1),除正常组ip给予等量的生理盐水外,其余各组在给药同时均ip D-半乳糖(125 mg.kg-1),连续给药6周。测定心、肝及皮肤等组织总抗氧化能力(T-AOC)、超氧化物歧化酶(T-SOD)活性、丙二醛(MDA)和羟脯氨酸(HYP)的含量。结果:与正常组比较,模型组肝、心和皮肤的T-SOD,T-AOC活性以及HYP含量均显著降低,差异显著(P<0.01),MDA含量明显增加,有显著性差异(P<0.01,P<0.05);与模型组比较,环黄芪醇能显著提高小鼠肝、心和皮肤的T-AOC,T-SOD活性(P<0.01),对肝、心和皮肤的MDA含量呈显著的降低作用(P<0.01,P<0.05),并可提高皮肤、心和肝等组织中HYP含量(P<0.01,P<0.05)。但环黄芪醇对上述指标的影响并未呈现明显的剂量依赖关系。结论:环黄芪醇具有显著的抗小鼠衰老作用,主要作用机制可能与提高T-SOD,T-AOC活性,降低MDA含量,增加HYP含量有关。 Objective:To investigate the effects and mechanism of cycloastragenol(CAG) on aging.Method:Mouse aging model was induced by D-galactose.Sixty mice were randomly divided into control group,model group,CAG(2.5,5.0,10.0 mg · kg-1)group and vitarmine E(VE,2.5 mg · kg-1)group.Each group was given ip D-galactose except normal control group was supplied with equal amount of physiological saline for 6 weeks,the treated mice were given medicines according dose.The activity of total antioxidant capacity(T-AOC) and total superoxide dismutase(T-SOD),the contents of malondialdehyde(MDA) and hydroxyproline(HYP) were mwasured.Result: Compared with the control group,the activity of T-SOD,T-AOC and the content of HYP in liver,heart and skin were markedly reduced in model group(P0.01),while the content of MDA was markedly increased(P0.01,P0.05);CAG group,compared with model group,significantly improved the activity of T-AOC in liver,heart and skin(P0.01).Meanwhile,the activity of T-SOD in liver,heart and skin(P0.01)and the content of HYP(P0.01,P0.05)were markedly increased.And it significantly reduced the content of MDA(P0.01,P0.05).Conclusions: CAG has a remarkable effect of anti-decrepitude,the possible mechanism is able to improve the activities of T-SOD,T-AOC and reduce the contents of MDA,HYP.
