摘要
目的:探讨滋补脾阴方药(Zibu Piyin Recipe,ZBPYR)对大鼠脊髓损伤(spinal cord injury,SCI)的治疗效果以及对低氧诱导因子(hypoxia inducible factor,HIF)-1α的表达的影响。方法:将清洁级Wistar大鼠36只随机分为假手术组、阳性对照组(喜得镇组,0.6 mg.kg-1)、SCI组和ZBPYR低、中、高剂量组(分别为7,14,21 g.kg-1)。各组大鼠分别于伤后72 h处死,并用Western-blot和RT-PCR方法检测HIF-1α和HIF-1αmRNA的表达。结果:伤后72 h,HIF-1α和HIF-1αmRNA在SCI组的表达量显著高于假手术组、ZBPYR低、中、高剂量组和阳性对照组,差异具有统计学意义(P<0.05);HIF-1α和HIF-1αmRNA的表达在ZBPYR低、中、高剂量组间的差异具有统计学意义(P<0.05)。结论:ZBPYR可改善受损脊髓局部微环境的缺血缺氧程度来抑制SCI后继发性损伤的发生和发展,其作用效果具有浓度依赖性。
Objective:To study the effect of Zibu Piyin Recipe(ZBPYR)on spinal cord injury and the influence on expression of hypoxia inducible factor(HIF)-1α.Method:Thirty-six adult Wistar rats were divided into six groups randomly: sham operated group,positive control group(hydergin,0.6 mg · kg-1),ZBPYR low,middle and high dose group(7,14,21 g · kg-1),SCI group.The rats were killed at 72 h post-injury.The expression of HIF-1α and HIF-1α mRNA was detected with RT-PCR and Western-blot assay.Result:At 72 h post-injury,the expression of HIF-1α and HIF-1α mRNA in SCI group were higher than that in the other groups(P0.05),and the expression of HIF-1α and HIF-1α mRNA in ZBPYR high,middle and low dose group were different statistically(P0.05).Conclusion:The ZBPYR could improve the function of neurons though improving oxygen-deficiency in local spinal cord tissue after injury.
