刺槐子叶的不定芽诱导及植株再生 被引量:4

摘要 以刺槐种子在无菌条件下播种生长7 d的子叶为外植体,研究不同植物生长调节剂和继代培养次数对愈伤组织诱导及植株再生的影响。结果表明:生长调节剂种类和质量浓度对刺槐子叶的愈伤组织诱导及愈伤组织再分化有一定影响,刺槐子叶诱导愈伤组织的最佳培养基:MS+2,4-D 1.0 mg/L+6-BA 0.2 mg/L,诱导率为98.9%,愈伤组织再生不定芽的最佳培养基:MS+6-BA 1.0 mg/L+NAA 0.05 mg/L,再生率为74.4%。愈伤组织的不定芽诱导率随继代次数的增加呈下降趋势。对刺槐子叶进行了直接诱导不定芽的试验,诱导率最高达87.8%,其培养基为:MS+6-BA 5.0 mg/L+2,4-D 0.1 mg/L。 The effects of different plant growth regulators and the number of subculture on callus induction and plant regeneration were studied by using seven-day-old cotyledons grown from Robinia pseudoacacia seeds as explants. Results showed that different types and concentrations of the plant growth regulators had certain influences on callus induction and adventitious bud regeneration from callus of black locust cotyledons. The best culture medium for inducing callus from cotyledon is MS supplemented with 1.0 mg/L 2,4-D and 0.2 mg/L 6-BA, with an induction rate of 98.9%. The optimal medium for ad- ventitious bud induction from callus is MS with 1.0 mg/L 6-BA and 0.05 mg/L NAA, with an induction rate of 74.4%. Adventitious bud induction frequency of callus declined as the subculture number increased. The highest induction rate of adventitious buds induced directly from the black locust cotyledons was 87.8%, and its best medium is MS with 5.0 mg/L 6-BA and 0.1 mg/L 2,4-D.
