肺纤方干预博莱霉素致肺纤维化大鼠的实验研究 被引量:7

Effect of Feixianfang on Pulmonary Fibrosis Model Induced by Bleocamicina in Rats

摘要 目的:观察肺纤方对博来霉素所致肺纤维化大鼠的影响。方法:144只健康Wistar大鼠随机分成空白组、模型组、阳性组、肺纤方高、中、低剂量组,每组各24只,按Szapiel法建立模型,次日ig给药,肺纤方高、中、低剂量组(3.2,1.6,0.8g.kg-1)均为10 mL.kg-1药液ig给药,阳性对照组用醋酸泼尼松混悬液(6 mg.kg-1)ig给药,空白组与模型组ig给予等量生理盐水,1次/d,给药周期21 d。分别在造模7,14,21 d后分3次取材,前、后期各随机杀检10只,中期杀检4只对照验证。腹主动脉取血检测细胞因子;解剖肺脏,右肺称重计算脏器指数后匀浆、离心、取上清液检测抗氧化酶系。结果:与模型对照组相比,肺纤方高剂量组在改善肺系数、升高谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)、降低丙二醛(MDA)、一氧化氮(NO)、转化生长因子(TGF-β1)、血小板衍生生长因子(PDGF)和肿瘤坏死因子(TNF-α)等方面差异具有统计学意义(P<0.05,P<0.01);中、低剂量组在不同方面几乎与醋酸泼尼松组作用相当;醋酸泼尼松组先期作用好于治疗组,但中、后期,肺纤方高剂量组的疗效明显好于醋酸泼尼松组,呈现明显的量效关系。结论:肺纤方是防治肺纤维化的一种有效方剂,对博莱霉素致大鼠肺损伤具有一定的保护作用,对博莱霉素所致的大鼠肺纤维化具有干预作用,且呈现一定的量效关系。 Objective: To observe the curative effect of Feixianfang on pulmonary fibrosis of rat model induced by bleocamicina. Method: One hundred and forty four healthy rats were randomly divided into six groups: normal group, model group, positive control group, Feixainfang high dose, middle dose and low dose group, 24 rats each group. The model were made with Szapiel's method, and the next day all rats were given the corresponding drugs with intragastric administration. The three treatment groups were given to high, middle and low dose of Feixianfang (3.2, 1.6, 0. 8 g kg-1 ), positive control group was given to prednisone acetate (6 mg kg-1). And normal group and model group were given normal saline, once a day, and the treatment lasted 21 days. Lungs were collected at the 7th day ( 10 rats), 14th day (4 rats), and 21s' day ( 10 rats) respectively. Result: Compared with model group, high dose treatment group showed significant difference in improving lung coefficient, increasing glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) and decreasing malondialdehyde (MDA) , nitrogen oxide (NO) , transforming growth factor-/31 ( TGF-/31 ) , platelet derived growth factor (PDGF) and tumor necrosis factor-a (TNF-a) (P 〈 0.05 or P 〈 0.01 ). Middle and low dose treatment group had the same effect as positive control group. And positive control group was better firstly than treatment group, but worse in intermediate stage and later stage. This directed that high dose treatment group had better effect than positive control group markedly, and the long-term effect was superior to prednisone acetate. Conclusion: Feixianfang is an effective formula in treating and preventing pulmonary fibrosis. It can intervene in the injury caused by bleocamicina with dose-effect relationship.
