摘要
选取栽培性状优良、较有代表性的 4个条斑紫菜栽培品系 ,将其自由丝状体采用RAPD标记技术进行分子标记和遗传结构的初步研究 ,用 2 8个随机引物在 4个品系中共扩增出 2 1 5条DNA片段 ,其中 2 3个引物扩增出的DNA片段图谱在 4个品系中是特异性的 ,可以用于品系鉴定 .RAPD图谱的统计结果表明 ,这 4个栽培品系间存在较大的遗传差异 ,而且在单孢子形成能力上差异较大者 ,其遗传距离也较远 .
The cultivar discrimination of laver(Porphyra yezensis)was examined with ran dom amplified polymorphic DNA(RAPD) analysis using purified genomic DNA.A total of 200 primers were used for the preliminary screening of RAPD primers,28 of whi c h produced distinct DNA bands from all of the 4 laver cultivars.Only one of thes e 28 showed identical patterns among the 4 cultivars while the remaining 27 prim ers showed polymorphic patterns.23 of the 27 primers produced polymorphic bands specific for a single cultivar.The genetic distances among the 4 cultivars were far (0.69~1.19).The genetic distances between cultivars which producese few monospores and which produces most monospores are far than that between any oth er two cultivars,which may mean that monospore production is an heritable chara cter.
