应用热乙醇法提取浮游植物中叶绿素a的探讨 被引量:10

摘要 [目的]利用热乙醇法提取浮游植物中叶绿素a。[方法]对叶绿素a的测定方法进行改进,通过对比试验探寻热乙醇法提取叶绿素a的最佳温度和反应时间,同时与现行规范法进行比较。[结果]用热乙醇法提取浮游植物中的叶绿素a,操作简便,安全可靠,萃取温度为75℃时,用热乙醇加热3 min提取时,叶绿素a提取效率最高。规范法和热乙醇法提取的叶绿素a存在显著正相关,热乙醇法且比规范法提取效率更高。[结论]该研究为地表水浮游植物中叶绿素a的提取提供了理论参考。 [Objective]The study aimed to extract chlorophyll a from phytoplankton by heat-ethanol method.[Method] The determination method of chlorophyll a was improved,the best extraction temperature and time of heat-ethanol method were explored,and then compared with the common conventional method.[Result] The heat-ethanol method is simple and safe to extract chlorophyll a,the chlorophyll a was the highest when extracted at 75℃ and then heated by hot ethanol for 3 min.There was a positive correlation of the chlorophyll a content between conventional method and heat-ethanol method,but the extraction rate by heat-ethanol method was higher than that by conventional method.[Conclusion] The study provided theoretical references for the extraction of chlorophyll a from phytoplankton.
