摘要
目的:研究人绒毛膜促性腺激素(HCG)联合免疫球蛋白对溴隐亭致大鼠流产模型蜕膜局部Th1/Th2型细胞因子及妊娠结局的影响。方法:采用孕第6、7、8天大鼠皮下注射溴隐亭0.3mg/kg,诱导SD大鼠流产模型,随机分为4组:即流产模型(A)组、HCG(B)组、HCG联合免疫球蛋白(IVIG)(C)组及正常妊娠(D)组,每组10只,妊娠第14天麻醉处死并观察妊娠结局,取胚胎吸收邻近的蜕膜研磨并裂解,离心后取上清液,-80℃保存。ELISA方法测定标本中肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)、白细胞介素-4(IL-4)以及白细胞介素-10(IL-10)的水平。结果:胚胎吸收率:A组明显高于其余3组,B组高于C、D组,差异均有统计学意义(P均<0.05)。Th1型细胞因子(TNF-α、IFN-γ)水平,A组明显高于其余3组,差异有统计学意义(P<0.05);Th2型细胞因子(IL-4、IL-10)水平,A组明显低于其余3组,差异均有统计学意义(P<0.05);IL-10细胞因子水平,B、D组低于C组,差异有统计学意义(P<0.05)。结论:HCG联合免疫球蛋白通过上调蜕膜局部细胞因子IL-10水平来改善胚胎发育,二者具有协同作用。
Objective.To investigate the effect of the combined therapy of HCG and human immunoglobulin G on local decidual Th1/Th2 type cytokines of bromocriptine induced aborting rats and its in- fluence on pregnancy's outcome. Methods: Rat models were induced by hypodermic injection of bromocrip- tine on the 6th ,7th ,8th day of pregnancy, and then divided into four groups: model group(group A), HCG group(group B), HCG and human immunoglobulin G group (group C); normal pregnant rats(D), On the 14th day after pregnancy, all rats were sacrificed to observe the fetes. Deciduas near the embryo were col- lected and mingled with the RIPA and PMSF, then grounded in a glass container for centrifugation, the su- pernatant were conserved at -80℃ for further use. The TNF-α. IFN-γ,IL-4 .IL-10's level of the supernatant were detected by ELISA method. Result:Fetal absorption rate of group A was significant higher than the other three groups(PG0.05) , the rate of group B was also significant higher than group B and C (P〈0. 05). Compared with group A, Th1 type cytokines (TNF-α, FN-γ,) were significantly decreased in group B, C and D (P〈0. 05) , while Th2 type cytokines (IL-4 and IL-10) all increased in the three groups(P〈0.5). Conclusions: The combined therapy of HCG and immunoglobulin G can improve the development of fetus by up-regulating local deeiduas IL-10 levels.
