人参皂苷Rd对Na_2S_2O_4致人脐静脉内皮细胞缺氧损伤的保护作用 被引量:2

摘要 目的建立Na2S2O4致人脐静脉内皮细胞(HUVEC)缺氧损伤模型,并观察人参皂苷Rd的保护作用。方法体外培养HUVEC,实验分为7组:空白对照组、溶媒对照组、连二亚硫酸钠(Na2S2O4)缺氧损伤组(模型组)、人参皂苷Rd不同浓度组(25μmol.L-1、125μmol.L-1、625μmol.L-1)、阳性药组(Nimodipine 50μmol.L-1),药物干预24 h后,除空白对照组和溶媒对照组外,其余各组用8 mmol.L-1Na2S2O4诱导HUVEC损伤4 h,进行相关指标检测:①倒置显微镜下观察细胞形态。②线粒体琥珀酸脱氢酶(MTT)法检测细胞存活率。③光化学法检测培养液LDH外漏浓度。结果 8 mmol.L-1的Na2S2O4缺氧损伤4 h可明显降低HUVEC的增殖活性,增加LDH的外漏,与空白对照组比较有显著性差异(P<0.01),表明HUVEC Na2S2O4缺氧损伤模型成功;人参皂苷Rd能明显减轻缺氧所致细胞形态学的改变、减轻内皮细胞损伤,提高HUVEC的MTT吸光度值,减少LDH的外漏,与模型组比较有显著性差异(P<0.01或P<0.05)。结论人参皂苷Rd对Na2S2O4致缺氧损伤的HUVEC具有保护作用,能提高细胞的存活率及改善其形态。 Objective To investigate the preventive effect of Ginsenoside Rd on the injury of human umbilical vein endothelial cells(HUVECs)induced by sodium dithionite(Na2S2O4).Methods HUVECs were cultured in vitro and injured by Na2S2O4(8 mmol·L-1 for 4 hours).They were divided into control group,solvent group,Na2S2O4 injury group,different final concentration of ginsenoside Rd(25 μmol·L-1、125 μmol·L-1、625 μmol·L-1)group,positive drug group(Nimodipine 50 μmol·L-1),in which the cells were injured by Na2S2O4 for 4 hours after they were incubated with different dosages of Ginsenoside Rd.① Cell morphology was observed by inverted microscope.② Cell viability was measured by MTT assay.③ The cytotoxicity of Na2S2O4 in HUVECs was determined based on the release of LDH into the incubation medium.The assay was in accordance with the manufacturer's instructions.Results 8 mmol·L-1 Na2S2O4 significantly reduced the absorbance values of HUVEC,increased LDH release,and compared with the control group,it was statistically significant.That showed the model was successed.In our experiment Ginsenoside Rd at different dosages could improve the absorbance values of HUVEC,reduce the release of LDH,compared with model group,it was statistically significant.Conclusion Ginsenoside Rd has a protective action on Na2S2O4-induced injury in HUVECs.It can improve the survival of endothelial cells activity and improve their shape.
