冷冻保护剂二甲基亚砜的毒副作用及应用剂量研究

Studies on toxicity and clinical application of cryoprotective agent dimethyl sulfoxide (DMSO)

本文主要用台盼兰排斥试验、CFU-GM体外琼脂培养技术和细胞电泳技术研究了冷冻保护剂二甲基亚砜(DNISO)在4℃和室温条件下对细胞活力的影响。结果表明,DMSO对骨髓、胎肝和淋巴细胞的毒性在室温比在4℃时大。小鼠静脉注射DMSO的LDS0剂量为3.9364±0.8709mg/g体重。提出了应用DMSO作为冷冻保护剂冻存人骨髓和胎肝细胞悬液的临床输注方法和DMSO的剂量安全值。

The effects of DMSO on the viability of bone marrow cells, fetal liver cells and lymphocytes at 4℃ and room temperature, the LD_(50) dose of DMSO in mice and dosage of DMSO used in patients are reported. It was shown that in comparison with the preservation at 4℃, cell viability and CFU-GM yield were markedly decreased when preserved at room temperature The LD_(50) dose of DMSO by single intravenous injection in mice was 3.0364±0.8709mg/g body weight. Our experimental studies in mice and dogs, and clinical application, revealed that unwashed and diluted cryopreserved bone marrow and fetal liver cell suspension may be successfully infused to animals and patients without any harmful side-effect.
