锌指蛋白(ZNF)185在小鼠不同发育阶段睾丸中的表达 被引量:4

Expression of ZNF185 in Different Developmental Stages of Mouse Testis

摘要 目的:研究锌指蛋白(ZNF)185在小鼠不同发育阶段睾丸中的表达。方法:从小鼠脑、肝、卵巢和不同发育阶段睾丸组织中提取总RNA,采用半定量RT-PCR法检测ZNF185的表达;制备小鼠不同发育阶段睾丸冰冻切片,应用免疫细胞化学分析法检测ZNF185蛋白的定位。结果:①ZNF185在睾丸中的表达显著高于脑、肝和卵巢组织。②ZNF185在小鼠性成熟前组睾丸中的表达显著低于性成熟期和老年期,而性成熟期和老年期之间差异不显著。③在性成熟前,ZNF185主要定位于睾丸间质细胞和类肌细胞;在性成熟期和老年期小鼠中,ZNF185主要定位于睾丸间质细胞、类肌细胞和精子。结论:ZNF185在小鼠性成熟后睾丸中的表达升高,定位更广泛。 Objective: To study the expression of ZNF185 in different developmental stages of mouse testis. Methods: Total RNA was extracted from mouse brain, liver, ovarian, and testicular tissue in different developmental stages, the expression of ZNF185 was detected by using semi-quantitative RT-PCR assay; the frozen sections of different developmental stages of mouse testis were made, the positioning of ZNF185 was detected using the immunocytochemical analysis. Results: 1) The expression of ZNF 185 in the testis was significantly higher than that in the brain, liver and ovarian tissues. 2) The expression of ZNF 185 in the testes of sexually mature before was significantly lower than that in the sexually mature group and age group, and there was no significant difference between sexually mature group and age group. 3) In the sexually mature before group, ZNF185 mainly located in the leydig cells and myoid cells. In sexually mature group and age group, the ZNF185 mainly located in the Leydig ceils, myoid cells and sperm. Conclusion: The expression of ZNF185 is increased in mouse testis after sexually mature, while the positioning is more widely.
