免疫亲和柱—高效液相色谱法检测乳制品中的磺胺

Rapid Method for the Determination of Sulfonamides in Milk and Powder Milk by Immunoaffinity Column Clean-up and HPLC-UV

建立了牛奶和奶粉中16种磺胺残留物的高效液相色谱—紫外快速测定方法。样品用乙酸乙酯提取,所得清液经免疫亲和柱净化富集,流动相为甲醇和1.1%乙酸—0.01 mol PBS溶液(梯度洗脱),应用液相色谱—紫外法检测。该方法的回收率为80%～110%,相对标准偏差小于10%;方法检测牛奶样品的灵敏度为0.5μg/L,奶粉样品的灵敏度为5.0μg/kg。

A high performance liquid chromatography-UV method is established for the determination of 16 sulfonamides residues in milk and powder milk clean up by immunoaffinity column （IAC） . After extraction with ethyl acetate, the clear part is loaded on the IAC column for purification, the mobile phase is methanol-1.1% acetic acid-0.01 mol PBS with gradient elution, and the chromatographic analysis is performed by HPLC. The recoveries of the method are in the range of 80%- 110%, the RSD is below 10%, the limit of quantification for milk and powder milk is 0.5 ug/L and 5.0 ug/kg, respectively.
