摘要
PDZ结构域在生物体中广泛存在,介导着大量的蛋白质相互作用,参与和执行多种生物过程和生理功能。PDZ结构域常规地结合配体蛋白C末端4~5个氨基酸残基。然而,蛋白质的相互作用可能比想象中复杂得多。研究发现PDZ结构域还存在着非常规的结合特性,PDZ结构域非常规结合的特性研究有助于丰富蛋白质相互作用网络,发现蛋白质新功能,并能以结合特性为基础研发安全有效的药物应用于相关疾病的治疗。
PDZ domains are popular model of protein interaction which mediate a variety of biological processes and physiological functions.They recognize canonical four to five amino acids at the C-termini of partner proteins.However,there are probably more patterns of protein interaction than we currently know.More and more studies showed that PDZ domains also have non-canonical binding specificities.Researches on the non-canonical binding specificities of PDZ domains can help to enrich the protein interaction networks and to discover novel functions of proteins,that may facilitate the development of safe and effective medicines for disease treatment.
