shRNA沉默RYBP基因表达对HL-60细胞增殖和凋亡的影响 被引量:1

摘要 Ring和YY1结合蛋白(RYBP)是生物进化过程中高度保守的转录抑制因子,目前认为是多梳(Polycombgroup,PcG)蛋白家族成员,与白血病、淋巴瘤、宫颈癌等肿瘤的发生、发展和预后密切相关。我们在前期实验中发现白血病患者骨髓单个核细胞(MNC)和白血病细胞系HL-60、SHI.1和K562细胞明显地高表达RYBP,且慢性髓性白血病(CML)患者治疗达到完全缓解时骨髓MNCRYBP表达量明显下降,复发时再升高。说明RYBP基因的表达异常与白血病的发生、预后等有关。我们试图通过慢病毒介导的RNA干扰技术沉默HL-60细胞RYBP表达,观察HL-60细胞增殖与凋亡,进一步探讨RYBP在白血病发生发展中的作用。
