茶树菇多糖联合化疗对食管癌大鼠TNF-α和IFN-γ的影响 被引量:6

Effect of Agrocyb aegerita polysaeeharide combined with chemotherapy on TNF-α and IFN-γ level in rat esophageal carcinoma model

摘要 目的:观察茶树菇多糖联合化疗对食管癌大鼠肿瘤坏死因子-α(TNF-α)和γ-干扰素(IFN-γ)的影响。方法:选用40只大鼠随机分为正常(正常对照)组,模型(模型对照)组,化疗(用替加氟)组,联合(替加氟联合茶树菇多糖)组。除正常组大鼠外,其他组大鼠皮下注射甲基戊基亚硝胺5 mg/kg,诱发食管癌模型,建模成功后各组治疗4周,记录大鼠的体重变化。用酶联免疫吸附(ELISA)法检测大鼠血清TNF-α和IFN-γ水平,采用免疫组化检测大鼠食管组织的TNF-α和IFN-γ表达情况。结果:在实验过程中,正常组大鼠体重呈逐步上升趋势,联合组大鼠体重呈缓慢上升趋势;而模型组和化疗组大鼠的体重变化不明显。联合组大鼠血清TNF-α水平明显低于模型组和化疗组,但高于正常组(P<0.05);而联合组大鼠血清IFN-γ水平明显高于模型组和化疗组,而低于正常组(P<0.05)。联合组大鼠的TNF-α阳性表达率低于模型组和化疗组,高于正常组;而IFN-γ阳性表达率高于模型组和化疗组,低于正常组(P<0.05)。结论:茶树菇多糖联合化疗可调节食管癌大鼠的免疫功能。其机制可能与下调TNF-α水平和上调IFN-γ水平有关。 Objective: The aim of this study was to observe the effect of Agrocyb aegerita polysaeeharide combined with chemotherapy on the levels of tumor necrosis factor-α(TNF-α) and Interferon-γ(IFN-γ) in rat model with esophageal carcinoma.Methods: Healthy Wistar rats were randomly divided into the following groups: normal control group(normal group),model control group(model group),Tegafur tablets group(chemotherapy group),and Tegafur tablets combined with Agrocyb aegerita polysaeeharide group(combination group).Except from the rats in normal group,the rats in other group were treated with MANA 5 mg/kg by subcutaneous injection to induce esophageal carcinoma model.The weight of rats among groups was recorded after 4 weeks treatment.The serum levels of TNF-α and IFN-γ in rats was detected using ELISA and the expression of TNF-α and IFN-γ was detected by immunohistochemistry method.Results: The serum TNF-α level in combination group was lower than that in model and chemotherapy group,and higher than that in normal group(P0.05).The serum IFN-γ level in combination group was higher than that in model and chemotherapy group,and lower than that in normal group(P0.05).The positive rates of TNF-α expression was lower than that in model and chemotherapy group,and higher than that in normal group.TNF-α expression was higher than that in model and chemotherapy group,and lower than that in normal group(P0.05).Conclusion: Agrocyb aegerita polysaeeharide combined with chemotherapy can regulate the immunological function in rat model with esophageal carcinoma,and the probable mechanism has correlation with down-regulation of TNF-α level and up-regulation of IFN-γ level.
