摘要
目的探讨神经干细胞(neural stem cells,NSC)移植治疗帕金森病的临床疗效。方法借助立体定向技术将原代培养的NSC(1×107/0.25 ml)缓慢注入临床确诊的12例帕金森病病人的尾状核头,移植前给予轻度热凝毁损。比较手术前后统一帕金森病评分量表(unified Parkinson's disease rating scale,UPDRS)评分。结果所有病人均顺利完成手术。帕金森病病人移植术后3个月临床症状的改善率为75%,术后6个月改善率为83.3%。所有病人随访12~24个月,平均18个月,改善率达75%。结论应用体外扩增的NSC可以安全、有效地治疗帕金森病,但远期疗效需进一步观察。
Objective To explore the clinical efficacy of neural stem cell (NSC) transplantation for Parkinson's disease (PD). Methods Twelve patients were confirmed as having PD by clinical diagnosis and primarily cultured NSCs (1 ~ 107/0.25 ml) were slowly injected into the head of the candate nucleus of patients by stereotaetie technique. Slight electrocoagulation lesion was performed in the patients before transplantation. The unified Parkinson's disease rating scale (UPDRS) before and after surgery were compared. Results All the patients underwent surgery successfully. The rates in symptom improvement were 75% and 83.3% 3 months and 6 months after transplantation respectively. All the patients were followed up for 12 to 24 months with a mean period of 18 months and the overall improvement rate was 75%. Condusions Administration of in vitro expanded NSCs is safe and effective for PD, but the long-term effectiveness needs to be further observed.
