摘要
目的:建立高效液相色谱法测定牛蒡子中绿原酸的含量,为绿原酸的药用植物资源开发提供依据。方法:依利特C18色谱柱(5μm,4.6mm×250mm),流动相为乙腈-0.4%磷酸(10∶90),流速:1.0 mL.min-1;检测波长:327nm;柱温:30℃。结果表明:绿原酸在2.82μg.mL-1~56.40μg.mL-1范围内与峰面积呈良好线性关系r=0.9999。平均回收率为100.2%,RSD=0.325%(n=6)。结论:该方法简便、准确、灵敏度高、重现性好,为牛蒡子的内在质量控制提供实验依据。
Objective: To determine the content of Chlorogenic Acid in Great Burdock Achene by HPLC for developing the medici- nal resources of Chlorogenic Acid. Methods: An external standard method by HPLC with Discovery YILITE Cls column (250 mm × 4.6 mm,25 μm) was employed. The mobile phase was consisted of acetonitrile -0.4% phosphoric acid solution (10 : 90) , at the flow rate of 1.0mL ~ min-I, the detection wavelength was 327 nm,column temperature was 30~C. Results: It showed good line- arity in the range of 2.82 - 56.4p^g ~ mL - 1 ( r = 0.9999). The content of Chlorogenic Acid in Great Burdock Achene was 0. 343% , and the average recovery rate of Chlorogenic Acid was 100.2%, RSD = 0. 325% ( n = 6). Conclusion: The method is simple, accu- rate, sensitive and reproducible. Internal quality control of the Great Burdock Achene is provided as experimental evidence.
