摘要
目的探讨NS-398对胃癌细胞侵袭能力的影响及可能机制。方法通过体外培养人胃腺癌细胞BGC-823,观察选择性环氧化酶2(COX-2)抑制剂NS-398(50、100、200μmol/L)对胃癌细胞迁移能力及侵袭相关分子基质金属蛋白酶2(MMP-2)、尿激酶型纤溶酶原激活物(u-PA)mRNA表达的影响。应用Millicell小室分析NS-398干预前后胃癌细胞迁移能力的变化;逆转录聚合酶链反应(RT-PCR)及Western blot检测NS-398干预前后胃癌细胞BGC-823的MMP-2、u-PA mRNA及其蛋白的表达变化。结果与干预前比较,NS-398干预后,胃癌细胞穿过微孔膜的细胞数明显降低,MMP-2、u-PA mRNA及其蛋白表达减少(P<0.05)。结论 NS-398在肿瘤的迁移和侵袭过程中发挥抑制作用,可能通过抑制MMP-2和u-PA mRNA转录和蛋白质合成,进而抑制肿瘤细胞外基质和血管基质降解而实现的。
Objective To explore the effects of NS-398 on cell migration and invasion of human gastric cancer cell line BGC-823. Methods Gastric cancer Cell lines were treated with NS-398. The mRNA and protein expressions of MMP-2 and u-PA were detected by RT-PCR and Western-blot, respectively. The invasion of cells treated with NS-398 was detected by Millicell study. Results Compated to before intervention, the number of the cells penestrating microhole membrane was reduced and the mRNA and protein expressions of MMP-2 and u-PA were decreased(P〈0. 05). Conclusion NS-398 can inhibit the cell migration and invasion of human gastric cancer cell line BGC-823,whieh may be related to its inhibiting effects on the mRNA and protein expressions of MMP-2 and u-PA.
