姜黄素对前列腺癌PC-3细胞增殖的抑制作用 被引量:4

摘要 目的研究姜黄素对PC-3细胞增殖活性的影响。方法以不同浓度(10、20、40、60、80μmol/L)的姜黄素及顺铂作用于体外培养的PC-3细胞24h,以中效浓度(40μmol/L)的姜黄素及顺铂作用于体外培养的PC-3细胞4、12、24、48、72h,后用MTT法检测在不同浓度及时间作用下PC-3细胞的抑制率,应用流式细胞仪检测PC-3细胞周期的变化。结果姜黄素对PC-3细胞的增殖抑制作用与顺铂相似(P>0.05),具有剂量时间依赖性,并随着药物浓度及时间的增加,抑制率逐渐增高,流式细胞仪检测显示:姜黄素和顺铂组都能使PC-3细胞生长明显阻滞于G2/M期,两组比较差异无统计学意义(P>0.05)。各实验组与空白对照组比较均有显著性差异(P<0.05)。结论姜黄素可以明显抑制PC-3细胞的增殖,在临床治疗前列腺癌方面有着良好的应用前景。 Objective To study the inhibitory effect of curcumine on prostate cancer cell line PC-3.Methods By means of different concentrations(10,20,40,60,80μmol/L)of curcumin and Platinol role PC-3 cells for 24 h,by means of lente concentration(40μmol/L) of curcumin and Platinol role PC-3 cells for 4,12,24,48,72 h.MTT method was used to measure the proliferation of PC-3 cells;and detect the cell cycle changes by flow cytometry.Results The depressant effect of curcumine on PC-3 is similar to Platinol(P 〉 0.05),with a concentration and time-dependent.As the drug concentration and time increases,the inhibition rate increased gradually.Flow cytometry showed that,curcumine and Platinol make PC-3 cell growth arrest at G2/M phase,there was no significant difference between the two groups(P 〉 0.05).Each experimental group compared with the control group had significant difference(P 〈 0.05).Conclusion Curcumine can significantly inhibit the proliferation of PC-3 cells and has a better value of clinical application and exploitation in the treatment of prostate cance.
