DNA甲基转移酶在肝细胞癌中的表达及其临床意义 被引量:1

摘要 目的:探讨DNA甲基转移酶1(DNMT1)、DNA甲基转移酶3a(DNMT3a)和甲基转移酶3b(DNMT3b)在肝细胞癌(HCC)中的表达及其意义。方法:应用免疫组化检测47例HCC癌组织、42例HCC癌旁肝组织及12例正常肝组织中DNMT1、DNMT3a和DNMT3b的表达情况,分析三者与临床病理特征之间的关系。结果:DNMT1、DNMT3a和DNMT3b在HCC中阳性表达分别为80.9%、68.1%和78.7%,在癌旁组织内的表达分别为50.0%、52.4%和57.1%,均明显高于正常肝组织内的阳性表达(16.7%、16.7%和25.0%),且DNMT1、DNMT3a和DNMT3b与HCC的病理分化类型及乙肝表面抗原阳性显著相关(P<0.05)。结论:DNMT1、DNMT3a和DNMT3b的异常表达与HCC的发生发展有紧密的关系。 Objective To investigate the expression and significance of DNA methyhransferase 1 (DNMT1), 3a(DNMT3a) and 3b(DNMT3b) in human hepatocellular careinoma(HCC). Methods The expression of DN- MT1,DNMT3a and DNMT3b was detected in 47 cases of HCC tumor specimen and 42 cases of HCC paracarci- noma liver tissue by immunohistochemistry, 12 cases of normal liver tissue as control; the results and clinical pathological parameters were analyzed. Results The positive expression rate of DNMT1, DNMT3a and DN- MT3b in HCC tissues was 80.9% , 68.1% and 78.7% respectively. The positive expression rate of DNMT1, DN- MT3a and DNMT3b in paracarcinoma tissues was 50.0%,52.4%and 57.1% respectively .All of them are signifi- cantly higher than that in normal liver tissues (16.7%, 16.7%和125.0% ). The expression of DNMT1,DNMT3a and DNMT3b was correlated with tumor differentiationand and hepatitis B surface antigen(HBsAg) (P 〈 0.05). Conclusion DNMT1,DNMT3a and DNMT3b maybe play important roles on carcinogenesis and the development of HCC.
