摘要
目的探讨类肝素结合表皮生长因子(heparin-binding epidermal growth factor-like growth factor,HB-EGF)的表达与超声分型预测子宫内膜容受性的关系。方法在排卵日B超对子宫内膜分型,分为A型组和C型组,于排卵后7 d手术获取两组的子宫内膜,C型组在取材的同时对子宫内膜进行局部机械刺激,第二周期继续观察排卵情况并在排卵后7 d时再次刮取子宫内膜归为SC型组。采用实时荧光定量逆转录聚合酶链反应(realtime-RT-PCR)、免疫组织化学方法测定HB-EGF在不同类型和机械刺激后子宫内膜中的基因表达水平和蛋向表达定位。结果 A型组和C型组人着床窗口期子宫内膜进行比较及与机械刺激前后子宫内膜(SC型)的比较时,HB-EGF的mRNA的表达和蛋白的表达在A型组高于C型组,并且经过机械刺激后SC型组HB-EGF的表达升高(P<0.05)。结论 HB-EGF与超声分型评估子宫内膜容受性正相关。
Objective: To explore the relationship between heparin-binding epidermal growth factor-like growth factor (HB-EGF) expression and ultrasonic pattern for evaluating the receptivity of human endometrium. Methods: The patterns of endomitrium were divided to pattern A, B and C according to the evaluation of ultrasonic examination on the ovulation day. The samples of pattern A and pattern C endomitrium were collected 7 days after ovulation in the spontaneous menstrual cycle. The endometrium was locally injured while the sample was taken from the patients with pattern C endomitrium. In the second spontaneous menstrual cycle, endometrial samples were collected again from the patient with pattern C endomitrium. The mRNA and protein expressions of HB-EGF were detected in different patterns of endometrium by real time-PCR and immunohistochemistry respectively. Results: The mRNA and protein expressions of HB-EGF in pattern A endometrium were significantly higher than those in pattern C endometrium in the window of implantation (P〈0.05). After local injury, the mRNA and protein expressions of HB-EGF was significantly increased (P〈0.05). Conclusions: HB-EGF expression was related to the classification of the ultrasonic patterns and could predict the outcome of implantation.
