摘要
生防细菌SF-16为土壤中筛选得到的1株细菌,研究表明,其对多种植物致病真菌有抑制作用。为了提高生防细菌SF-16的芽胞形成率及芽胞数量,以及为工厂化生产生物菌剂提供理论依据,采用摇瓶发酵对芽胞形成的主要影响因素进行了考察。通过单因素实验和正交实验对SF-16菌株产芽胞条件进行了分析,确定了摇瓶发酵最佳条件为:乳糖0.5%、黄豆饼粉2.0%、MnSO4·5H2O0.30%。在该优化条件下,发酵液中颉颃菌的出芽率达到98.3%,最终生物量达到2×109个/mL。
Biocontrol bacteria SF-16 was a selected bacteria strain,research showed that its inhibitory effect on multiple plants.In order to improve biocontrol bacteria SF-16 spore formation rate and quantity,provide theoretical basis for factory production of biological agent,use flask fermentation with a survey on main influencing factors of spore formation.Through single factor experiment and orthogonal experiment,the optimal shaking flask fermentation condition was determined as follow:media composed of 0.5% lactose,2.0% soybean meal,0.30% MnSO4·5H2O.Under this optimum condition,the germination rate achieves 98.3 percentages,Biomass achieves 2×109/mL.
