红花多糖对人胃癌SGC-7901细胞抑制作用的初步研究 被引量:6

摘要 目的观察红花多糖(SPS)对人胃癌SGC-7901细胞增殖与凋亡的影响。方法不同浓度的SPS相同时间作用于人胃癌SGC-7901细胞,采用倒置显微镜观察SGC-7901细胞的形态学变化及增殖情况,吖啶橙染色观察SGC-7901细胞的凋亡情况。结果 SPS处理组贴壁细胞的数量明显减少,细胞体积变小,部分细胞崩解。吖啶橙染色结果显示,经SPS处理组细胞胞质浓缩、染色增强,部分细胞核固缩为新月状。结论 SPS能抑制人胃癌SGC-7901细胞体外生长,同时可诱导SGC-7901细胞凋亡,该作用呈一定的时间与剂量依赖。 Objective To observe the effect of Safflower polysaccharide (SPS) on the cell proliferation and apoptosis of human gastric carcinoma cell line SGC-7901. Methods SGC-7901 cells were processed with different concentrations of SPS during the same period. The morphology changes and proliferation of SGC-7901 cells were observed wiht inverted microscope. The apoptosis of SGC-7901 cell was detected by acridine orange(AO) assay. Results SGC-7901 cells in SPS group showed bad refraction, shrink, and more dead cells in culture medium were observed. Results of AO fluorescent staining showed part of SGC-7901 cells of cytoplasm condensed,chromatin increased, and the crescent-shaped shrinkage could be seen in some cells nucleus. Conclusion SPS can markedly inhibit the growth of SGC-7901 cells and induce apoptosis of gaatric carcinoma cell line SGC-7901 in vitro cultured in a time-and dose-dependent manner.
