摘要
目的探讨缺氧诱导因子-2α(HIF-2α)对结肠癌细胞SW480增殖的影响。方法常规培养结肠癌细胞SW480,采用CoCl2处理细胞24 h制备细胞缺氧模型,通过绘制细胞生长曲线观察细胞生长速度和倍增时间的改变;通过流式细胞术观察细胞周期分布的改变。结果缺氧+RNA干扰组HIF-2αmRNA及蛋白的表达与单纯CoCl2处理组相比均明显降低(P均<0.05);单纯缺氧组与对照组、缺氧+siRNA1组和缺氧+siRNA2组相比,细胞生长速度明显加快,倍增时间显著缩短(P均<0.05),而对照组与缺氧+RNA干扰组相比生长速度和倍增时间无明显差异(P>0.05);单纯缺氧组与对照组和缺氧+RNA干扰组相比,未出现明显的G1期阻滞,S期和G2/M期细胞比例显著增加(P<0.05),缺氧+RNA干扰组与对照组相比无显著性变化(P>0.05)。结论 HIF-2α可促进结肠癌细胞SW480增值;靶向HIF-2α的siRNA能够抑制结肠癌细胞SW480中HIF-2α的表达,并抑制SW480细胞的增殖能力。
Objective It is to approach the influence of hypoxia inducible factor-2α(HIF-2α) on the proliferation of colorectal cancer cells SW480.Methods Colorectal cancer cells SW480 were routine cultured to prepare hypoxia model by CoCl2,then the changes of cell growth rate and doubling time were observed by cell growth curve,the changes of cell cycle were observed by flow cytometry.Results The expressions of HIF-2α mRNA and protein in hypoxia+RNA interference groups were significantly lower than that in pure CoCl2 group(P0.05).Compared with control group and hypoxia+RNA interference groups,cell growth speed of simple hypoxia group was significantly accelerated,doubling time was significantly shortened(P0.05).Compared with control group and hypoxia+RNA interference groups,there was no obvious G1 phase arrest in simple hypoxia group,the proportion of S phase and G2/M phase cells were increased significantly(P0.05).Conclusion HIF-2α can promote the proliferation of breast cancer cell SW480;HIF-2α targeted siRNA can inhibit the expression of HIF-2α and the proliferation of colorectal cancer cells SW480.
