摘要
[目的]应用RAPD技术分析甲型溶血性链球菌DNA多态性并对其分型。[方法]随机设计17hp和10hp 两条引物,采用近年来建立起来的RAPD技术对30株甲型溶血性链球菌DNA进行扩增,扩增产物经凝胶电泳后得到 指纹图。[结果]共出现12个DNA条带,根据指纹图的异同将其分为1、R、1型别。[结论]甲型溶血性链球 菌 DNA用 RAPD技术分析,把该菌进一步分型且方法操作简便,结果准确、稳定。
: [Objective ]: To analyse the DNA polymorphism and identify the strain types in a-hemolytic streptococcus by using Randomly Amplified pdymorphic DNA. [Methods ] Randomly designing 17hp and 10hp oligonucleotide as primers. We used the technique of Randomly Amplified pdymorphic DNA,'which has been developed in recent years, to amplify the DNA of 30 strains of a-hemolytic streptococcus and followed the agarose electrophoresis of the amplified products and the finger prints were revealed. [Result ] Altogether there were 12 scorable markers. The 30 strains were differentiated into I ~ Ⅲ type. [Conclusions ] RAPD is a kind of accurate method to analyze a-hemolytic streptococcus and also a reliable molecular biological method because of the simplified operation and the stable results.
