三苯氧胺对人卵巢癌细胞增殖和凋亡的影响 被引量:2

摘要 目的:探讨三苯氧胺的不同剂量(0.1、1、10μmol/L)对人卵巢癌细胞增殖和凋亡的影响,为卵巢癌的内分泌治疗提供实验依据。方法:以体外培养人卵巢癌细胞系HO-8910为研究对象,用免疫组化SABC法检测增殖细胞核抗原(PCNA)在细胞中表达情况,DNA缺口原位末端标记方法(TUNEL)检测细胞凋亡。结果:三苯氧胺通过诱导细胞凋亡抑制卵巢癌细胞生长,至剂量依赖性。低浓度(≤1μmol/L)的三苯氧胺对PCNA表达的影响无统计学意义,高剂量(10μmol/L)可明显降低PCNA表达。结论:三苯氧胺抗肿瘤作用与剂量有关,低剂量通过诱导细胞凋亡发挥作用,而高剂量与抑制细胞增殖和诱导凋亡有关。 Objective: To investigate the effect of tamoxifen on proliferation and apoptosis of human ovarian cancer cell and provide some experimental data to the endocrinal therapy of ovarian carcinoma. Methods: Human ovarian cancer cell line HO - 8910 was studied. The expression of proliferation cell nuclear antigen (PCNA) of HO - 8910 was detected with immunohistochemical staining. Apoptotic index(AI) was detected using DNA in situ terminal deoxynucleotidyl transferase - mediated dUTP - biotin nick ending labeling(TUNEL). Results: Tamox ifen inhibited proliferation and induced apoptosis in a dose - dependent to human ovarian cancer cell. Tamoxifen in low doses (≤1μmol/L) inhibited the growth of ovarian cancer via apop tosis and not alter PCNA expression (P > 0.05 ), but in high dose (10μmol/L), not only induced apoptosis but also reduced PCNA expression (P <0. 05 ). Conclusions: The mechanism of tamoxifen antitumor is linked to its doses.Tamoxifen in low doses inhibits proliferation of ovarian cancer cell via apoptosis, in high dose inhibition associated with poliferation - inhlbited and induction apoptosis.
