摘要
目的:建立黄芪党参颗粒中黄芪甲苷含量的HPLC测定方法。方法:以ELSD为检测器,色谱柱C18(Alltech Hypersil ODS 4.6 mm×150 mm,5μm);流动相:乙腈∶水(40∶60),流速:1.0 mL/min;柱温:25℃;漂移管温度:100℃,载气量:2.6 L/min。结果:回归方程y=1.5471x+2.6231(r=0.9996),黄芪甲苷在1.656～4.968μg/mL之间峰面积与浓度线性关系良好。平均回收率为101%。结论:该法简便可靠,可为黄芪党参颗粒质量控制提供依据。
Objective： To establish a method for content determination of astragaloside Ⅳ in HuangQi DangShen granule with HPLC.Method： ELSD was taken as detector,chromatographic column was C18（Alltech Hypersil ODS 4.6 mm×150 mm,5 μm）;mobile phase： methyl cyanide-water（40∶60）,flow rate： 1.0 mL/min;column temperature： 25℃;drift tube temperature： 100℃,containing gas 2.6 L/min.Result： The regression equation was y= 1.5471x＋ 2.6231（r=0.9996）.The excellent linear ranges was 1.656 ～4.968 μg/mL.Average recovery rate was 101%. Conclusion： The method,simple and reliable,could provide evidence for quality control of HuangQi DangShen granule.
