摘要
目的探讨硫酸酯茯苓多糖(S—PCS3-Ⅱ)诱导胃腺癌细胞凋亡的作用。方法体外培养低分化胃腺癌细胞株MKN-45,加入不同浓度的硫酸酯茯苓多糖作用于MKN-5细胞不同作用时间后,利用细胞形态学、流式细胞术(FCM)、DNA琼脂糖凝胶电泳、吖啶橙/溴化乙锭双染法、透射电镜等实验方法观察对细胞凋亡的影响。结果硫酸酯茯苓多糖作用MKN45细胞后,细胞形态学观察见抑制细胞生长,浓度为0.5g/L的硫酸酯茯苓多糖作用24h、48h、72h后其凋亡率分别为(7.92±0.83)%、(20.68±1.75)%、(37.52±2.47)%,且DNA凝胶电泳出现梯状条带。荧光显微镜和电镜下可见典型的细胞凋亡形态变化。结论硫酸酯茯苓多糖可诱导胃腺癌细胞凋亡来抑制胃腺癌细胞生长。
Objective To investigate the effect of the sulfated derivatives polysaccharides from sclerotium of Poria cocos ( S - PCS3 - II) on gastric adenoearcinoma MKN - 45 cells apoptosis. Methods The MKN - 45 cell line was cultured in vitro, and was exposed to a range of concentrations of S - PCS3 - II at different time. The effect of apoptosis was observed by cell morphology, flow eytometry, agarose gel DNA electrophoresis, fluorescent microscopy and transmission electronmicroseopy. Results S - PCS3 - II can inhibit the growth of human gastric cancer MKN -45 cells. When MKN -45 cells were treated for 24 h ,48 h ,72 h at 0.5 g/L concentration, the apoptosis rate revealed (7.92 ± 0.83 ) %, (20.68 ± 1.75 ) %, ( 37.52 ± 2.47 ) %, respectively. DNA agarose gel electrophoresis showed the typical DNA ladder of apoptosis. A typical apoptosis morphology was also observed by fluorescent microscopy. A typical apoptosis ultrastructure changes was also observed by transmission electronmicroscopy. Conclusion S - PCS3 - II can inhibit the growth of human gastric cancer MKN - 45cells by inducing gastric cancer cells apoptosis.
