摘要
利用荧光光谱法研究了1,4,7,10-四氮杂环十二烷合铜(Cyclen-Cu(Ⅱ))配合物与牛血清白蛋白(BSA)的相互作用,用Stern-Volmer方程、Lineweaver-Burk方程确定了其荧光猝灭机理为静态猝灭,得到1,4,7,10-四氮杂环十二烷合铜配合物和BSA作用的结合常数、结合点位数,计算了在不同的pH值、温度及表面活性剂溶液中的热力学参数,实验结果表明其作用力主要为静电作用力.还探讨了表面活性剂对Cyclen-Cu(Ⅱ)与BSA作用的影响,实验结果显示表面活性剂使得BSA在348nm处的荧光峰蓝移约12nm并且其强度随表面活性剂浓度的增加而降低.
The interaction of cyclen-Cu(Ⅱ) with bovine serum albumin(BSA) was investigated with fluorescence spectroscopy.The results showed that Cyclen-Cu(Ⅱ) strongly quenched fluorescence of BSA.Using the Stern-Volmer equation and the Lineweaver-Burk equation,it was proved that the fluorescence quenching mechanism belonged to the static quenching.According to the Lineweaver-Burk equation and the thermodynamics equation,the binding constants,binding states and thermodynamics parameters were obtained at different temperatures in the aqueous solution and the surfactant solutions.The electrostatic force played a main role in the binding of Cyclen-Cu(Ⅱ) with BSA.The wavelength of the fluorescence spectrum apexes changed to shortwave at 348 nm in the surfactant solutions.
