加味四逆散对慢性应激所致大鼠雌激素紊乱的调节作用 被引量:1

Regulation Effects of Jiawei Sini San on Chronic Stress-induced Estrogen Disorders in Rats

摘要 目的:研究加味四逆散对慢性不可预计性刺激致抑郁雌性大鼠血清皮质酮(Cort)、血清雌二醇(E2)的影响,揭示其对雌性大鼠抑郁模型内分泌紊乱的调节机制。方法:Wistar大鼠按体重和基础糖水消耗量随机分为6组:空白组(不施加任何刺激)、慢性轻度不可预计应激模型组、加味四逆散高、中、低剂量组(315,158,78 mg.kg-1,ig),阳性药氟西汀组(18 mg.kg-1,ig),应激第1天开始每天8:30 ig给药,正常组及慢性应激模型组每次ig蒸馏水。应激第22天取材。放射免疫法检测用药前后雌性大鼠血清Cort,E2的含量变化。结果:大鼠血清Cort空白组(0.05±0.04)μg.L-1,模型组为(0.11±0.08)μg.L-1,较空白组显著升高(P<0.05),加味四逆散各组Cort较模型组低但差异无统计学意义;血清E2含量模型组大鼠的E2水平(0.04±0.02)μg.L-1较空白组(0.12±0.03)μg.L-1明显下降(P<0.05),加味四逆散高、中、低剂量组E2依次为(0.10±0.04),(0.08±0.02),(0.07±0.05)μg.L-1均明显升高,与模型组相比有显著性差异(P<0.01,P<0.05)。结论:加味四逆散抗抑郁作用与其维持E2正常分泌有关。 Objective: To observe influence of Jiawei Sini San on serum corticosterone (Cort) , estradiol (E2 ) content in chronic mild unpredictable stress rats, and reveal the adjustment mechanism of endocrine disorders in female rat depression. Method: Wistar rats were randomly divided into six groups according to the weight and basis for sucrose : control group, model group, Jiawei Sin San high, medium and low dose group (315, 158, 78 mg· kg^(-1), ig), positive drug fluoxetinegroup (18 mg·kg^(-1), ig), stress with every day 8:30 ig, was performed on the first day control group and model group were ig given distilled water. On 22 day of stress, samples were collected to measure serum Cort, serum E2 content changes by radioimmunoassay in female rats. Result: Serum Cort concent in model group (0. 11 ± 0.08) μg·L^(-1), compared with the control group (0.05 ± 0.04) μg ·L^(-1) was significantly increased (P 〈 0.05 ). In Jiawei Sini San group eort concentration was lower than in model group, but the difference was not statistically significant. In model group serum E2 levels (0. 04 ± 0.02) μg ·L^(-1) compared with control group (0. 12 ± 0.03) μg·L^(-1) decreased significantly (P 〈 0.05 ). but compared with model group was not significant. In Jiawei Sini San group high, medium and low dose group serum E2 levels (0. 10 ±0.04), (0.08 ±0.02), (0.07 ±0.05) μg ·L^(-1) were significantly elevated, increased compared with the model group (P 〈0.01, P 〈0.05). Conclusion: The antidepressant effect of of Jiawei Sini San may be related to secretion of the E2.
