琥珀酸合用利培酮抗精神疾病研究

摘要 目的探讨琥珀酸(succinate)的抗精神疾病作用。方法①在大鼠杏仁核电刺激点燃模型上灌胃给药琥珀酸0.1,0.4,0.8 g.kg-1,30 min后测定后放电时程和Racine’s分级并与给药前比较。②56例住院精神分裂症患者随机分为两组,分别进行利培酮合并琥珀酸治疗(研究组)和单独的利培酮治疗(对照组),并分别于治疗前和治疗后8周末采用PANSS量表评定药物疗效,TESS量表评定药物不良反应的情况。结果①琥珀酸灌胃给药(0.8 g.kg-1),在大鼠杏仁核电刺激点燃模型上,可缩短点燃后放电时程ADD(P<0.05),降低Racine’s分级(P<0.01);②治疗8周末,研究组和对照组PANSS总分、兴奋因子分的变化均有显著性差异(P<0.05)。两组间不良反应的发生率和嗜睡的发生率存在显著性差异(P<0.05),其他各不良反应发生率无显著性差异。结论琥珀酸对利培酮治疗精神分裂症有明显的减副增效作用。 Objective: To investigate the antipsychotic effect of succinate.Methods: ① Succinate(0.1,0.4,0.8g·kg-1) was administrated by gavage to amygdala electrical kindled rats,and afterdischarge duration(ADD) and Racine's score were determined 30min later and compared them with those before gavage.② Fifty-six cases of hospitalized schizophrenic patients were randomly divided into two groups,with one group treated with risperidone alone and the other group combined with succinate.Before and after treatment in 8 weeks PANSS scale was used to assess drug efficacy and TESS scale assess adverse drug reactions.Results: ① Succinate(0.8g·kg-1) significantly shortened ADD(P0.05),lower Racine's score on amygdala electrical kindled rats(P0.01).② After 8-weeks treatment,PANSS total score and exciting subscore of the study group were significantly lower than those of the control group(P0.05),and the incidence of adverse reactions and sleepiness of the study group were significant lower than those of the control group(P0.05),the incidence of other adverse reactions had no significant difference.Conclusion: For the treatment of schizophrenia,succinate can increase the efficiency and reduce Vice synergies of risperidone.
