摘要
目的研究子痫前期胎盘组织氧化应激情况及巨噬细胞移动抑制因子(MIF)的表达,探讨MIF在子痫前期氧化应激中的作用。方法选择2010年3~10月在青岛大学医学院附属医院住院分娩的孕妇71例,其中子痫前期孕妇38例(子痫前期组),同期行剖宫产的正常晚期孕妇33例(对照组)。采用逆转录RT-PCR技术检测各组孕妇胎盘组织中MIF mRNA的表达,测定各组胎盘组织中超氧化物歧化酶(SOD)活力及丙二醛(MDA)、过氧化氢(H2O2)含量。并对对照组及子痫前期组孕妇胎盘组织中H2O2含量与MIF mRNA表达的相关性进行分析。结果 (1)子痫前期组MIF mRNA的表达(1.12±0.14)明显高于对照组(0.90±0.12)(P<0.01)。(2)子痫前期组SOD[(86.15±17.53)U/mgprot]明显低于对照组[(101.00±21.00)U/mgprot](P<0.01);MDA[(153.74±28.47)nmol/mgprot]明显高于对照组[(100.63±14.25)nmol/mgprot](P<0.05);H2 O2[(112.68±21.73)nmol/mgprot]明显高于对照组[(94.64±18.68)nmol/mgprot](P<0.01)。(3)对照组胎盘组织中H2O2与MIF mRNA表达水平呈正相关关系(r=0.65,P<0.01);子痫前期组胎盘组织中H2O2与MIF mRNA表达水平呈正相关关系(r=0.74,P<0.01)。结论子痫前期患者胎盘组织中MIF的过度表达,引起血管内皮细胞损伤,造成氧化应激状态,从而参与子痫前期的发病。
Objective: To investigate the role of macrophage migration inhibition factor(MIF) and oxidative stress in placenta of preeclampsia.Methods:Seventy-one women were recruited,including 38 preeclampsia women and 33 healthy pregnant women as the control group.The expression of MIF mRNA in placenta was determined by Semiquantitative RT-PCR.The homogenates of placenta were estracted and the acitifity of superoxide dismutase(SOD) and the quantity of malondialdehyde(MDA),hydrogen peroxide(H2O2) were measured.The relationship between the level of H2O2 and MIF mRNA in placenta was analyzed in the control group and the preeclampsia group.Results:(1)The expression of MIF mRNA in preeclampsia(1.12±0.14)was significantly higher than that in the control group(0.90±0.12)(P0.01).(2)The activity of SOD of homogenate of placenta in preeclampsia was 86.15±17.53 U/mgprot,which was lower than that of control 101.00±21.00 U/mgprot(P0.01),while MDA was 153.74±28.47 nmol/mgprot,which was higher than that of control 100.63±14.25 nmol/mgprot(P0.01).H2O2 in preeclampsia with the quantity 112.68±21.73 nmol/mgprot was higher than that of control 94.64±18.68 nmol/mgprot(P0.01).(3)Positive correlation between H2O2 and MIF mRNA was found both in the study(r=0.74,P﹤0.01) and the control group(r=0.65,P﹤0.01).Conclusion:Over expression of MIF in the placenta can result in the vascular endothelium damage and oxidative stress which may be involved in the pathogenesis of preeclampsia.
