大豆苷元对异丙肾上腺素所致心肌肥厚的保护作用

摘要 目的建立异丙肾上腺素(1so)诱导的大鼠心肌肥厚模型,观察大豆苷元(DD)对大鼠心肌肥厚时抗氧化功能的影响。方法健康雄性sD大鼠随机分为5组:对照组、心肌肥厚模型组、溶剂对照组、低剂量0.1m01.kg-1及高剂量0.3m01.kg-1DD组。各组分别背部皮下注射同体积Ns或异丙肾上腺素1mg.kg-1,每日一次,连续10d,建立心肌肥厚模型。连续皮下给药14d。通过测定全心重量参数、左心室重量指数、心肌组织羟脯氨酸含量、血清MDA含量和SOD活性来判断DD对心肌肥厚的保护作用。结果模型组和溶剂对照组的全心重量参数、左心室重量指数及心肌组织羟脯氨酸含量明显高于对照组(P〈0.001),与模型组和溶剂对照组相比,DD组的全心重量参数(P〈0.01)、左心室重量指数(P〈0.05)及心肌组织羟脯氨酸含量降低(P〈0.001);模型组和溶剂对照组的MDA含量明显高于对照组(P〈0.001),而不同浓度DD组则显著性降低;高剂量DD组的SOD活性较模型组高(P〈0.05);高、低剂量DD组的血清GSH—PX活性均较模型组高(P〈0.001)结论DD对异丙肾上腺素诱导的大鼠心肌肥厚具有保护作用,提高大鼠的抗氧化功能是DD抗心肌肥厚的机制之一。 Objective To explore daidzein' s (DD)anti-oxidation effect on rat' s myocardial hypertrophy induced by isopropylarterenol (Iso). Methods The rat's myocardial hypertrophy model was formed through hypodermic injection which extended 10 days at back skin with Iso (1 mg.kg-1.d), and was administrated with DD (0.1 or 0.3 umol.kg-1.d), DMSO or NS in intragastric way from the second day, and extend to 14 days. After the last administration, the rat was put in absolute diet status for 12 hours and its blood and heart was taken under anesthesia. The myocardial hypertrophy index expressed on heart weight/body weight (HW/BW) and left ventricle weight/body weight (LVW/BW). The concentration of MDA and hydroxyproline and the activities of SOD and GSH-PX in serum were analyzed. Results It demonstrated that HW/BW and LVW/BW of Iso group were much bigger than the control group, and the concentration of MDA in serum and hydroxyproline in myoeardium were also enhanced. Compared with Iso group, DD groups reflected decreases in the index, hydroxyprolin and MDA level, and increases in the activities of SOD and GSH-Px. Conclusion DD reflected a protective effect on Iso-induced myocardial hypertrophy in rat. It may be related to reducing the oxidative stress and improving antioxidant capacity.
