壬基酚对雄性大鼠体重、脏器指数以及睾丸超微结构的影响

摘要 目的初步研究不同剂量壬基酚对雄性大鼠的毒性作用。方法雄性大鼠48只,随机分为4组,即对照组、壬基酚高(200mg·kg-1)、中(100mg·kg-1)、低剂量组(50mg·kg-1),给药组每日灌胃给予壬基酚油溶液,连续28d;对照组每日灌胃给予植物油(ig,0.15ml/100g),连续28d。之后处死大鼠,迅速摘除睾丸、附睾、精囊、脾脏、胸腺、肾上腺和肾脏,称重并计算脏器指数。另外,摘取大鼠右侧睾丸,锋利预冷刀片切开,取1mm×1mm×2mm的组织于戊二醛磷酸缓冲液固定,超薄切片,透射电镜观察睾丸超微结构变化。结果与对照组相比,给予壬基酚后大鼠体重增长程度受抑,高剂量组(200mg·kg-1)体重增长幅度最小(P〈0.05)。染毒组大鼠睾丸、附睾、精囊指数下降,高剂量组大鼠精囊指数显著降低(P〈0.05);大鼠肾脏指数增加,肾上腺、脾脏、胸腺和肾脏指数没有变化。电镜下大鼠睾丸生精细胞排列出现紊乱,细胞器变性多见;支持细胞线粒体空泡化,嵴消失,内质网肿胀。结论壬基酚减轻大鼠体重,降低生殖相关脏器指数,导致大鼠睾丸超微结构损伤。 Objective To study the toxic effects of various doses of nonylphenol (NP) on the male rats. Methods 48 SI) male rats were randomly divided into four groups, control group, high-dose NPgroup (200 mg kg-1), middle-dose NP group (100 mg.kg-1), low-dose NP group (50 mg. kg-1). Then the rats were perfusing a stomach with nonylphenols oil solution for 28 days continuously while the control group was given vegetable oil. 28 days later, all the rats were executed, and their testis, epididymis, seminal vesicle, spleen, thymus, adrenal gland and kidney were removed, and the organ indexes were calculated after weighing. In addition, the rates' right testis was removed quickly, fixed in phosphate buffer solution of glutaraldehyde. Then ultra-thin section was made, and the uhrastructure was observed under electron microscope. Results Compared with control group, body weight gains of the nonylphenol rats were decreased significantly and the weight gains of high-dose group (200 mg, kg-1) was the least (P〈0.05). The organ index of testis, epididymis and seminal vesicle were decreased, and the seminal vesicle index of high-dose group were decreased significantly (P〈0.05). And the kidney index of nonylphenol rats was increased with no changes found in the organ index of adrenal gland, thymus and spleen. Conclusion The body weight and the organ index related to reproduction were decreased; the ultrastructure of rat's testis was damaged by nonylphenol.
